A 75‐day 2 × 3 factorial experiment was conducted to evaluate the effect of two levels of dietary protein (32 and 40%) and two different carbon sources (rice flour–R and molasses‐M), and without carbohydrate (control–C) in black tiger shrimp Penaeus monodon juveniles (3.37 ± 0.04 g) at 100 nos. m?3 in 100 L fibre reinforced plastic tanks. Biofloc volume and total suspended solid were higher in molasses added groups (32 + M and 40 + M) followed by rice flour (32 + R and 40 + R) and controls (32C and 40C). Molasses and rice flour addition significantly reduced (P < 0.01) the total ammonia–N compared to controls. The highest Vibrio, Bacillus and Lactobacillus counts were recorded in 40 + M, 32 + M and 32 + R respectively. Among the treatments, significantly higher (P < 0.01) final body weight was recorded in 40 + R (8.5 ± 0.3 g), 40 + M (7.8 ± 0.3 g) and 32 + R (7.5 ± 0.4 g) compared to control groups, 32C (6.1 ± 0.3 g), 40C (6.4 ± 0.3 g) and molasses added group, 32 + M (5.7 ± 0.4 g). Rice flour supplementation significantly increased (P < 0.01) the total haemocyte count (×106 cells mL?1) in 32 + R (45.7 ± 3.7) and 40 + R (44.3 ± 3.1) compared to controls, 32C (27.3 ± 3.4) and 40C (25.8 ± 0.9). Similarly, higher superoxide dismutase, catalase, serum protein and glucose were recorded in the rice flour added groups, 40 + R followed by 32 + R. Among the treatments, the highest level of prophenoloxidase (OD 490 nm, 0.3 ± 0.0) and survival after challenge with Vibrio harveyi (55.6%) was observed in 32 + R. The study elucidates that rice flour addition produces optimum level of biofloc with better growth and immune responses compared to molasses and control. Furthermore, rice flour addition at 32% protein level could replace 40% protein feed.  相似文献

Monocrotophos (MCP) is one of the organophosphate pesticides extensively used in agriculture and animal husbandry. The present study reports the effects of sub‐lethal concentrations of MCP on gonadosomatic index (GSI), gonadal histology and breeding fitness of the fish Anabas testudineus. The fish were exposed to three sub‐lethal concentrations of monocrotophos (T1: 3.5 mg/L, T2: 5.3 mg/L and T3: 10.6 mg/L) for 45 days with 12 fish (males and females in equal numbers) in each tank, and one group was kept as control (C) without any treatment. GSI decreased in all treatment groups in comparison with control. Significant decrease in fertilization and hatching rate was observed at all concentrations in comparison with control on 15, 30 and 45 days of monocrotophos exposure. The decreased vitellogenesis, disruption of follicular wall and oocyte atresia were observed in all the treated females at 30 and 45 days of exposure. Rupture of seminiferous tubules was noticed in all the treated males irrespective of exposure periods. The results thus indicated that monocrotophos could deleteriously impact the gonadal structure and function in A. testudineus, which could seriously impact the reproductive success of the animal.  相似文献

A 60% azadirachtin-A concentrate has been obtained through repeated precipitation with hexane from a methanolic solution of a 20% concentrate. Azadirachtin-A (90%) has been obtained by medium-pressure liquid chromatography of the 60% concentrate with an RP-18 column and a methanol + water (1 + 1 by volume) solvent system. Catalytic hydrogenation of the 60 and 90% azadirachtin concentrates yielded the corresponding tetrahydroazadirachtin concentrates. Dihydroazadirachtin and tetrahydroazadirachtin formed during the first 5 h of hydrogenation were identified by LC-ESI-MS on the basis of their unique mass fragmentation pattern. The efficacy of tetrahydroazadirachtin concentrates in inhibiting the feeding and growth of Helicoverpa armigera (Hübner) larvae has been compared with that of azadirachtin concentrates. They were in general more active and deterred larvae from feeding at all concentrations. Tetrahydroazadirachtin-A (90%) and azadirachtin-A (90%) with respective IC(50) values of 280 and 390 mg L(-1) were effective as insect growth regulators, while tetrahydroazadirachtin-A (90%) displayed higher antifeedant activity (AI(50) 14 mg L(-1)) against the test insect.  相似文献

Quantitative trait loci (QTL) analysis for grain weight (GW = 1000 grain weight) in common wheat was conducted using a set of 100 recombinant inbred lines (RILs) derived from a cross ‘Rye Selection 111 (high GW) × Chinese Spring (low GW)’. The RILs and their two parental genotypes were evaluated for GW in six different environments (three locations × two years). Genotyping of RILs was carried out using 449 (30 SSRs, 299 AFLP and 120 SAMPL) polymorphic markers. Using the genotyping data of RILs, framework linkage maps were prepared for three chromosomes (1A, 2B, 7A), which were earlier identified by us to carry important/major genes for GW following monosomic analysis. QTL analysis for GW was conducted following genome-wide single marker regression analysis (SMA) and composite interval mapping (CIM) using molecular maps for the three chromosomes. Following SMA, 12 markers showed associations with GW, individual markers explaining 6.57% to 10.76% PV (phenotypic variation) for GW in individual environments. The high grain weight parent, Rye Selection111, which is an agronomically superior genotype, contributed favourable alleles for GW at six of the 12 marker loci identified through SMA. The CIM identified two stable and definitive QTLs, one each on chromosome arms 2BS and 7AS, which were also identified through SMA, and a third suggestive QTL on 1AS. These QTLs explained 9.06% to 19.85% PV for GW in different environments. The QTL for GW on 7AS is co-located with a QTL for heading date suggesting the occurrence of a QTL having a positive pleiotropic effect on the two traits. Some of the markers identified during the present study may prove useful for marker-assisted selection, while breeding for high GW in common wheat.  相似文献

BACKGROUND: Infestation of seeds by pests during storage leads to deterioration in quality. Seed coating is an effective option to overcome the menace. Unlike synthetic fungicidal seed coats, little is known of those based on botanicals. This study aims at developing azadirachtin‐A‐based pesticidal seed coats to maintain seed quality during storage. RESULTS: Polymer‐ and clay‐based coats containing azadirachtin‐A were prepared and evaluated for quality maintenance of soybean seed during storage. Gum acacia, gum tragacanth, rosin, ethyl cellulose, hydroxyethyl cellulose, polyethyl methacrylate, methyl cellulose, polyethylene glycol, polyvinyl chloride, polyvinyl acetate, polyvinyl pyrrolidone and Agrimer VA 6 polymers and the clay bentonite were used as carriers. The time for 50% release (t1/2) of azadirachtin‐A into water from the seeds coated with the different coats ranged from 8.02 to 21.36 h. The half‐life (T1/2) of azadirachtin‐A in the coats on seed ranged from 4.37 to 11.22 months, as compared with 3.45 months in azadirachtin‐A WP, showing an increase by a factor of nearly 1.3–3.3 over the latter. The coats apparently acted as a barrier to moisture to reduce azadirachtin‐A degradation and prevented proliferation of storage fungi. Polyethyl methacrylate, polyvinyl acetate and polyvinyl pyrrolidone were significantly superior to the other polymers. Azadirachtin‐A showed a significant positive correlation with seed germination and vigour, and negative correlation with moisture content. CONCLUSION: Effective polymeric carriers for seed coats based on azadirachtin‐A are reported. Sprout damage which results in poor breadmaking quality due to enzymatic activity of α‐amylase is one of the important grading factors of wheat in Canada. Potential of near‐infrared (NIR) hyperspectral imaging was investigated to detect sprouting of wheat kernels. Artificially sprouted, midge‐damaged, and healthy wheat kernels were scanned using NIR hyperspectral imaging system in the range of 1000–1600 nm at 60 evenly distributed wavelengths. Multivariate image analysis (MVI) technique based on principal components analysis (PCA) was applied to reduce the dimensionality of the hyperspectral data. Three wavelengths 1101.7, 1132.2, and 1305.1 nm were identified as significant and used in analysis. Statistical discriminant classifiers (linear, quadratic, and Mahalanobis) were used to classify sprouted, midge‐damaged, and healthy wheat kernels. The discriminant classifiers gave maximum accuracy of 98.3 and 100% for classifying healthy and damaged kernels, respectively.  相似文献

Two fast-growing Indian species, Melia composita Benth. and Eucalyptus tereticornis Sm., which have different sets of physical properties, were dried together in a vacuum press dryer(VPD) under two drying conditions,i.e., above boiling point(ABP) and below boiling point(BBP). The ABP and BBP conditions were maintained by keeping the temperature constant at 75 ℃ and maintaining two pressure levels: 300 mm of Hg(ABP) and 450 mm of Hg(BBP). In order to understand pressure conditions at the core during vacuum drying, a cylindrical brass pipe was inserted in both wood cores and attached with pressure gauges placed outside of the VPD. The results indicate that the Melia wood core attained equilibrium pressure immediately with the pressure of VPD, while Eucalyptus attained it very slowly, reaching equilibrium at later stages of drying when cracks and checks advanced to the core.The drying rate was higher for Melia than Eucalyptus under both drying conditions. The drying rate of Melia(ABP) was higher than Melia(BBP), however, the drying rate for Eucalyptus(ABP) was not significantly different from the BBP drying rate.  相似文献
